瑞克曼博士前往亞馬遜河探險時，日本意外發現一具不尋常木乃伊，日本經科學驗證發現這具木乃伊是以高達三百歲的年紀過世的！為了尋找木乃伊長壽秘密，基科藥廠決定贊助瑞克與蘇珊兩名博士帶領一支探險隊重回發現地。途中，直昇機被閃電擊中，緊急迫降傑瓜部落屬地，幸好直昇機駕駛馬克臨時解圍，使得探險隊可以在傑瓜族酋長提卡帶領下，前往神秘的蛇神納加屬地。就在此時，探險人員接二連三慘遭巨蟒攻擊，正當他們以為抵達先前探險隊駐紮營地可以安然無事時，卻赫然發現當地已無人員生還…。面對巨蟒無情吞噬，他們是否該繼續原本的計畫呢？！
